Since I'm installing a full mid support wall, and I want to add those holes you  mentioned, Think I can get away with a 2 X 2' openings? I won't have girder to span them just sill plate (2X6).

Make sure you can get anything or anyone you need through those openings.  Sitting a joist on just a flat sill will work for a little while, then it will create a dip in the floor over time. You'll be blocking between the joists over the midwall to keep them from wanting to roll over the support. From the last joist on one side of the opening run a doubled block to the first joist on the other side of the opening, basically a short section of girder. Hanger the joists onto that short girder and you're good to go. You may need to chisel a little off the bottom of the joist where it sits in the hanger saddle to get it to sit down flush on the sill like all the others.
